Depends on what type of joint and how much glue you have added...
If you are butt jointing and putting a few screws into the end-grain then yes it will hold - until you use it

2" thick timber and screws only 2 1/2" long?

If you're screwing through 2" thick timber you should really be using screws 3 1/2" long, or 4" if you are screwing into end grain.

Also, I've only used "yellow" wood glue once - the dog pee'd up the bottle

Ahh the problem here is the american PAR timbers. Ours comes up roughly 1/4 of an inch smaller than the sawn size. The usa timbers are a good half inch smaller.

So our timbers 4 x 2 = 3 3/4 x 1 3/4 inch or 95mm x 45mm
usa timbers are about 3.5'' x 1.5''

The recommended screw size is normally about 1.5 times? the thickness of the timber

But i would normally use 3 or 3.5 inch screws for joining 4 x 2 timbers or similar. :)
