You could apply a varnish or lacquer over the once dried painted area for extra protection and also use a decent exterior quality paint in the first place.

you can certainly rout it, but given that you are asking the question, i would suggest a lot of practicing on scrap first. It isnt the easiest of tasks and would be far easier with a sign writing template.

If you are painting in the letters and then using a glaze/varnish/lacquer to protect, do make sure paint and top-coat are compatible.
It's worth doing a test on some scrap to make sure.
